What makes two words one letter apart?

Two words are one letter apart when they are the same length and match in every position except one, like COLD and CORD, or WARD and WARM. Every rung in a solved climb connects to its neighbour by a single letter change.

Do the levels get harder?

Yes. Early levels use 4-letter words, then 5-letter, and the hardest use 6-letter words. Every ladder has a single correct ordering, so it is always solvable by logic, and a Reveal a letter nudge is there if you get stuck.
